The big kahuna of the city's improv theatres, UCB hosts shows featuring stars from Saturday Night Live, Late Nite With Conan O'Brien, etc, rubbing elbows with the stars of tomorrow. (Or perhaps the day after.)
Details and Ticketing
For the most part, these shows are just like any other play - there's a stageful of actors, dialogue, lights, an audience... only the script didn't exist before the play began. It's a little hit-and-miss - some shows are better than others. (For a first experience, you're best off going to one of the "featured" shows on the bill.)

The PIT (The 'insiders' fave.)
Gotham City Improv (Wildly varying fare, very annoying flash website, upstairs from a decent sushi place.)
Chicago City Limits (Funny, but institutional. We suspect they have pension plans.)
National Comedy Theatre (New kids on the block.)
